hnpccMy cells are still not grown enough for me to make competent cells out of them.
I'm debating with myself - do I wait around till (probably) 8pm or come in at 7am.
I've been waiting all day for them to grow. My protocol reckons they hit OD600 ~ 0.5 after overnight growth. Well so far they're at OD ~ 0.2, and they've been going for just over 24 hours. I'm tired. I'm sick. I'm sick of bloody agrobacterium.
I just want to make my competent cells, zap them with DNA and go home and sleep.
I wish I lived closer than 35km away from Monash. Then I could go home, eat, sleep, come back and make cells.
Argh.
I think I'll OD them again at 6 and see where they're at now... *sigh
